There was already a Mainz medical orchestra in the 1960s, at which time it consisted exclusively of members of the university's medical department. After a large number of the orchestra members were gradually appointed to other universities and clinics, the ensemble initially disbanded.

In 1981-82, the internists at the university hospital launched an initiative to reestablish a medical orchestra. Professors, assistants, and students from the medical department came together, and over the years, an ensemble developed that roughly corresponded to a typical small symphony orchestra. Gradually, the orchestra was supplemented by members from other departments, resulting in a colorful mix of students and staff from various departments of the university, as well as external members.
